Two reports have shown that mammalian artificial chromosomes (MAC) can be constructed from cloned human centromere DNA and telomere repeats, proving the principle that chromosomes can form from naked DNA molecules transfected into human cells. The MACs were mitotically stable, low copy number and bound antibodies associated with active centromeres. As a step toward second-generation MACs, yeast and bacterial cloning systems will have to be adapted to achieve large MAC constructs having a centromere, two telomeres, and genomic copies of mammalian (...) genes. Available construction techniques are discussed along with a new P1 artificial chromosome (PAC)-derived telomere vector (pTAT) that can be joined to other PACs in vitro, avoiding a cloning step during which large repetitive arrays often rearrange. The PAC system can be used as a route to further define the optimal DNA elements required for efficient MAC formation, to investigate the expression of genes on MACs, and possibly to develop efficient MAC-delivery protocols. MORRIS: But come back to that other kind of fiction, in which the author himself is involved with his works, not merely in writing something for other people but in writing what seems to be necessary to his conscious existence, to his sense of well-being. For such a writer, when he finished with something he finishes with it; he is not left with continuations that he can go on knitting until he runs out of yarn. This conceit reflects my own (...) experience as a writer, relying on the sap that keeps rising, the force that drives the flower, as Dylan Thomas put it. It is plantlike. We put it in the sun and when it doesn't grow, we take it and put it in another room. I don't think of repotting the plant. The plant must make its own way. BOOTH: I like the organic metaphor, but I keep wanting to come back to particular cases to see how you actually work, in literal detail. Even the organic novelist obviously still has the matter of collecting notes, starting a novel, having it fail to go. Let me put a simple question, and move out from there. How many actual novels, whether they ever reach fruition or not, do you have "growing" at a given time? MORRIS: You don't mean simultaneously? BOOTH: I mean actual notes that exist in some kind of manuscript form, starts on a novel, something you are actually working on. MORRIS: It is so unusual for me to have more than one or two things in mind at once that I don't find this a fruitful question. The history of contemporary modal logic dates back to the writings of C. S. Lewis in the early part of this century. Since then, a growing body of literature has attested to professional interest in the area, and in a number of related issues in philosophical logic which have received wide attention. The recent development of powerful formal techniques for modal system building, together with an increasing interest in modal logic as a tool for philosophical analysis, have created a need (...) for an up-to-date text to introduce students to this material. Snyder's book attempts to answer this need. Assuming an understanding of elementary propositional logic, Snyder introduces a reductive technique called cancellation for detecting the theorems of a system of logic. This technique, analogous to the construction of Smullyan trees but more compact, is extended to modal and quantified contexts. Cancellation versions of the systems M and Mn of G. H. von Wright, and S4 and S5 of Lewis are developed. Snyder uses a Hintikka type of semantics, showing how the various formal systems are differentiated at the semantic level by differing conditions which must be imposed on the model systems used as interpretations for them. These model systems, and the conditions imposed upon them, are in turn described by means of a metalanguage which is essentially first-order quantificational logic. The power of this technique stems from the fact that for every object language formula of a system, there is a corresponding formula in the metalanguage which describes the conditions an interpretation must meet in order to satisfy the original formula. The conditions that the interpretations of a given system of logic must meet are reflected in this metalanguage as "antecedent assumptions," and these in turn correspond to cancellation rules for the object language. This correspondence is constructed in such a way that the metalinguistic counterpart of each theorem of the system will be a theorem of first-order logic. Snyder's primary interest is in showing how modal logic can be used, with the help of these techniques, to build a large variety of formal systems and to "tailor" such systems to meet a variety of analytical tasks. Simple modal systems are developed for the articulation of a number of modal concepts, in addition to the classic alethic ones. Examples are taken from temporal, deontic, and epistemic logic to show how specific interpretations of the meanings of the relevant operators lead to the incorporation of appropriate conditions in the formal systems used to articulate them. An entire chapter is devoted to a discussion of the paradoxes of material and strict implication, and to an attempt to articulate the notion of entailment through the development of formal systems which include a dyadic modal operator that is free of these paradoxes. The final chapter discusses such classical issues as proper names, reference, fictional entities, definite descriptions, and existence presuppositions. Appendices deal with such matters as the equivalence of the cancellation systems to more classical axiomatic-deduction systems, and the sketch of a proof of soundness and completeness for all the modal systems presented. While designed as a text, this volume should be of interest to both logicians and those working in metaphysics and language analysis, since the primary concern of the author is to develop techniques that will facilitate the usefulness of modal logic as a tool for philosophical analysis. Suffering evokes moral and metaphysical reflection, the bioethics of suffering concerns the proper ethos of living with suffering. Because empirical and philosophical explorations of suffering are imprisoned in the world of immanent experience, they cannot reach to a transcendent meaning. Even if religious and other narratives concerning the meaning of suffering have no transcendent import, they can have aesthetic and moral significance. This understanding of narratives of suffering and of their custodians has substantial ecumenical implications: chaplains can function as general (...) custodians of narratives and sustainers of a generic religious meaning. This understanding is contrary to traditional Christianity, which discloses a transcendent significance of human suffering found in a very particular history involving particular persons: Christ as the second Adam through the submission of the second Eve has taken on our nature so that we can be united with God. Human suffering is tied to human sin, not simply as a punishment for sin, much less as an opportunity to discharge a supposed temporal punishment due to sin. Human suffering is the result of our rebellious free choices. It provides an opportunity for humility and submission, so that, united to the cross of Christ, sin can be forgiven and suffering set aside in the Resurrection. Knowledge of this framing context for all human suffering is accessible not through rational argument. It is a knowledge garnered through repentance, purification of the heart, illumination by God's grace, and unification with God. Christian bioethics is embedded in the narrative of suffering, which is part of the history of salvation and which encompasses and places all of medicine in its terms. (shrink)

Because sin is anything that turns our heart from God, sins are both voluntary and jnvoluntary. As a consequence, double effect can only be adequately understood in a Christian context in which it is recognized that, even when evil is not willed, our involvement in its causation can still mar our hearts. The acknowledgement of involuntary sins resituates double effect so that the traditional Christian concern with spiritual harm and healing can be maintained. In this way, one can overcome the (...) underdeveloped Western theological appreciation of double effect, which ignores the effect on the heart of causing evil. (shrink)

In the face of the moral pluralism that results from the death of God and the abandonment of a God's eye perspective in secular philosophy, bioethics arose in a context that renders it essentially incapable of giving answers to substantive moral questions, such as concerning the permissibility of abortion, human embryonic stem cell research, euthanasia, etc. Indeed, it is only when bioethics understands its own limitations and those of secular moral philosophy in general can it better appreciate those tasks that (...) it can actually usefully perform in both the clinical and academic setting. It is the task of this paper to understand and reevaluate bioethics by understanding these limits. Academic bioethicists can analyze ideas, concepts, and claims necessary to understanding the moral questions raised in health care, assessing the arguments related to these issues, and provide an understanding of the different moral perspectives on bioethical issues. In the clinical setting, bioethicists can provide legal advice, serve as experts on IRBs, mediating disputes, facilitating decision-making and risk management, and clarifying normative issues. However, understanding this is only possible when one understands the history, genesis, and foundations of bioethics and its inability to provide a resolution to postmodern moral pluralism. (shrink)

This paper presents four different understandings of the family and their concomitant views of the authority of the family in pediatric medical decision making. These different views are grounded in robustly developed, and conflicting, worldviews supported by disparate basic premises about the nature of morality. The traditional worldviews are often found within religious communities that embrace foundational metaphysical premises at odds with the commitments of the liberal account of the family dominant in the secular culture of the West. These disputes (...) are substantial and ultimately irresolvable by sound rational argument because of the failure to share common foundational premises and rules of evidence. It is in light of these fundamental disagreements that there is a need to evaluate critically the claims and agenda advanced by the Convention on the Rights of the Child. (shrink)
